Bioinformatic design of a multi-epitope protein derived from human transglutaminase with potential for serological diagnosis of celiac disease

Background and Aim: Celiac disease is the most common autoimmune disorder of the digestive tract, which is generally diagnosed in childhood and its global prevalence is increasing. Given that the existing serological diagnostic tests do not have very high sensitivity and specificity, there is a need to develop efficient serological diagnostic tests. The aim of the present study was the bioinformatic design of a multi-epitope protein construct derived from epitopes of human transglutaminase protein, which can be used in the next steps of the project as a recombinant protein to design and produce an ELISA test for more accurate serological diagnosis of celiac disease. Methods: In this project, the human transglutaminase antigen, which is used in existing serological tests, was selected and its effective epitopes were selected using bioinformatics methods and tools, and finally a multi-epitope protein structure was designed and its model was built. In the final stage, the stability of the designed protein was evaluated using advanced calculations and molecular dynamics simulation. Results: The designed structure was formed based on linear epitopes recognized by B cells. This structure had suitable physicochemical properties and the predicted 3D model for it was similar to natural proteins. Finally, the results of molecular dynamics simulations showed that this protein could have suitable stability. Conclusion: The multi-epitope antigen designed in this study can be used to produce ELISA kits for serological diagnosis of celiac disease using serum samples from affected children.
