The interaction between FTO gene polymorphism and dietary fat intake in male adolescents

Background and Aim: Childhood obesity, a global health crisis, is influenced by genetic and environmental factors. The FTO gene, a key regulator of body mass, interacts with dietary patterns, though evidence linking its polymorphisms to specific fat intake remains inconsistent. This study examines the association between the FTO rs9930506 polymorphism and dietary fat intake in male adolescents. Methods: A cross-sectional study was conducted with 222 male high school students from Tehran, Iran. Data on sociodemographic, nutrition (using a 168-item FFQ), physical activity (IPAQ), and anthropometrics were collected. Blood samples were analyzed for FTO rs9930506 genotypes via PCR sequencing. Statistical analyses included chi-square tests and linear regression (SPSS v23, α = 0.05). Results: Participants with AA genotypes (14.22 ± 3.11 years) exhibited higher body weight (p = 0.03) and lower caloric intake (p = 0.04) compared to AG/GG carriers (13.85 ± 1.00 years). No significant differences in physical activity were observed (AA: 1189.50 ± 1334.61 vs. AG/GG: 1123.55 ± 2146.94 MET-min/week, p = 0.76). AA genotypes showed a 1.8-fold increased likelihood of elevated saturated fat intake (95% CI: 1.2–2.7) and a reduced consumption of omega-3 fatty acids (β = -0.34, p = 0.01). Trans fat intake was correlated with higher BMI percentiles in AA carriers (r = 0.42, p < 0.001). Conclusion: The FTO rs9930506 polymorphism influences dietary fat preferences and adiposity markers in male adolescents, underscoring the gene-nutrient interaction in the pathogenesis of obesity. These findings support the use of genotype-tailored dietary interventions in the management of adolescent obesity.
